The arrival of the action potential causes the first influx of calcium ions, allowing the vesicle to dock at the active zone. But the fusion proteins are only activated by a second influx of calcium ions through additional channels at the zone. When the proteins fuse the vesicle to the presynaptic membrane, the vesicle opens and empties its neurotransmitters into the synaptic cleft.
